An HPLC purity figure is the main peak's share of total detected peak AREA — a statement about proportions, not about how much material is in the vial.

The separation, then the counting

High-performance liquid chromatography pushes a dissolved sample through a packed column. Different molecules travel at different speeds depending on how strongly they interact with the packing, so what entered as a mixture leaves as a sequence of separated components. A detector at the far end records each one as it emerges, producing a trace with a peak for every compound the instrument could resolve.

Why the result is a percentage of area, not of mass

The number quoted as purity is the area of the main peak divided by the total area of all peaks, expressed as a percentage. Area is used because the detector responds to concentration over time rather than to weight directly. This is the single most important thing to understand about the figure: it describes the proportions of what was detected, and says nothing whatever about how much material there was.

What it cannot see

Anything that does not absorb at the wavelength being monitored is invisible to the detector, and anything that does not travel through the column at all never reaches it. Salts, residual solvent and water commonly fall into one or other category. A 99% figure therefore means 99% of what this method could see, which is why a mass or content test is a separate measurement rather than a formality.

Why two labs can report different numbers

Column chemistry, gradient, wavelength and run length are all choices, and they change which impurities separate from the main peak and which hide underneath it. Two honest laboratories can return figures a few tenths apart on the same vial without either being wrong. This is also why comparing a purity figure from one lab against one from another is weaker evidence than comparing two reports from the same lab.

What to do with the number

Read it as a statement about composition from one method, dated, on one sample. Treat differences of a few tenths between reports as noise rather than signal. Treat a difference of several points as worth asking about. And treat the absence of a companion content or identity test as the real gap, because purity alone leaves the two most expensive failure modes — wrong compound, wrong quantity — unexamined.

Why the wavelength matters

The detector watches one wavelength, usually chosen because the target compound absorbs strongly there. That choice is a decision about what will be visible. An impurity that absorbs poorly at the chosen wavelength contributes a smaller peak than its actual share of the material, and one that does not absorb at all contributes nothing. The figure is therefore method-dependent in a way the single number conceals, which is a reason to prefer a long series from one laboratory over single readings from several.

Peaks that hide under peaks

Separation is never perfect. An impurity that travels at nearly the same speed as the target compound emerges at nearly the same time, and its peak can sit partly or wholly beneath the main one. Better methods separate more, which is why a longer run or a shallower gradient sometimes produces a lower purity figure on the same vial — not because the material changed, but because the measurement got better at seeing.

What a chromatogram shows that a number does not

The trace itself carries information the summary figure discards: how many impurities there were, whether they were one large one or many small ones, and whether the main peak is clean or shouldered. Reports that publish the trace are offering more than reports that publish only the percentage, and the difference costs the laboratory nothing to provide.
